Olaparib is a poly(ADP-ribose) polymerase (PARP) inhibitor, the first in its class, whose mechanism of action is based on the principle of synthetic lethality in cells with defective homologous recombination (e.g., in BRCA1/2 mutations). The drug is used for the treatment of various oncological diseases, including ovarian, breast (including in the adjuvant setting), pancreatic, and prostate cancers.
